How it works

5-Amino-1MQ (5-amino-1-methylquinolinium) is a small-molecule, membrane-permeable inhibitor of nicotinamide N-methyltransferase (NNMT), a cytosolic enzyme highly expressed in white adipose tissue and liver. It is a methylquinolinium compound, not a peptide despite its frequent grouping with research peptides. NNMT normally consumes nicotinamide and the universal methyl donor S-adenosylmethionine (SAM) to produce 1-methylnicotinamide (1-MNA) — a reaction researchers describe as a 'methyl sink' that can divert substrate away from NAD+ salvage.

By occupying the NNMT active site, 5-Amino-1MQ has been shown in preclinical work to lower intracellular 1-MNA while raising NAD+ and SAM and suppressing lipogenesis in adipocytes. In diet-induced obese mice it has been studied for reduced body weight and white adipose mass without changing food intake. It is not FDA-approved and is handled as a research compound; no published human clinical trials currently characterize its safety or efficacy.
